Background
PCSK2(Proprotein Convertase, Subtilisin/Kexin-Type, 2), also known as PC2 or NEC2, is an enzyme responsible for the first step in the maturation of many neuroendocrine peptides from their precursors, such as the conversion of proinsulin to insulin intermediates. Seidah et al.(1991) mapped the NEC2 gene to human chromosome 20p11.2-p11.1. Ohagi et al.(1992) observed expression of the chloramphenicol acetyltransferase reporter fusions containing the putative promoter region in insulinoma cells but not in hepatoma cells, consistent with the known tissue-specific pattern of expression of the PC2 gene. Analysis of the level of CAT activity with several deletion mutants identified the region from -1100 to -539 from the translation start site as essential for PC2 promoter activity.
